5-甲基吡嗪-2-羧酸是一种重要的医药中间体,主要用于合成第二代磺脲类降血糖药,新一代长效降血脂药和治疗结核病的有效药物。
本论文以 2,5-二甲基吡嗪为原料,高锰酸钾为氧化剂,经过一步氧化合成 5-甲基吡嗪-2-羧酸。该工艺反应进行到一定程度的时候就会达到平衡,原料 2,5-二甲基吡嗪未能全部参加反应,造成浪费。如若再增加高锰酸钾用量,则会使二氧化副反应产物明显增加。为使原料充分利用和提高收率,本论文研究了高锰酸钾用量和母液循环套用对产品收率和成本的影响,实验结果表明,在一定范围内, 2,5-二甲基吡嗪与高锰酸钾摩尔比和产率成正比,母液循环套用次数和产率成正比,且纯度已达到医药中间体的要求,有望实现工业化生产。

Abstract
5- methyl pyrazine -2- is an important medical intermediate, mainly used for the synthesis of the second generation sulfonylurea hypoglycemic agents, a new generation of long-term effective drug reducing blood lipid drugs and treatment of tuberculosis.
In this thesis the 2,5- two methyl pyrazine as raw material, Potassium Permanganate as oxidizing agent, after one step oxidation synthesis of 5- methyl pyrazine -2-. The process of reaction to a certain extent will reach equilibrium, the raw material 2,5- two methyl pyrazine failed to fully participate in the reaction, resulting in a waste of. If we add Potassium Permanganate dosage, will make the two oxidation side reaction products increased significantly. In order to make full use of raw materials and increase the yield, the paper research the Potassium Permanganate dosage and mother liquor circulation apply effects on product yield and cost, the experimental results show that, in a certain range, two 2,5- methyl pyrazine and Potassium Permanganate molar ratio and yield is proportional to, mother liquor circulation apply the number and yield is proportional to, and the purity reached pharmaceutical intermediates requirements, can realize the industrialization production.
