Abstract：Sister chromatid differentiation（SCD）is a chromosome processing technology developed in the mid 70s. The technology can be used to study a series of important theoretical issues，such as， the cell cycle, semiconservative replication,the molecular structure and aberrations of chromosome, as well as DNA replication, damage and repair.Besides,SCD can also be used for the analysis of sister chromatic exchange frequency.SCE is able to detect the change frequency of chromosome，and present a dose-response relationship,therefor,SEC is regard as one of the conventional indicators of detection of mutations and teratogenicity testing.In this study，we use a sample of grass and the SCD method is used to process samples,and then observe the morphology and color difference of sister chromatid under the microscope.

Abstract: sister chromatids dyeing method (SCD) distinction is the mid-seventies development of chromosome processing technology. This technique are used to study the cell cycle, chromosome duplication, chromosome half reserves molecular structure and distortion, and DNA damage and repair, and so on a series of important theoretical problem, also can be used to analyze the sister chromatids swap (SCE) frequency. Due to the sensitivity test chromosome SCE can change, showing doses, therefore, has caused the SCE as detecting mutant, one of the conventional index carcinogens. This experiment with human chromosomes specimens for experimental material, using the same method for specimen SCD, under the microscope sister chromatids of form and color.
