本实验以石刁柏花蕾为材料,采用石蜡切片法制片,观察了石刁柏花药的解剖结构,比较分析了石刁柏花蕾外部形态特征以及石蜡切片观察,得出二者之间对应关系;以石刁柏花药为材料,进行花药培养得出适合石刁柏花药培养的诱导培养基、分化培养基及培养条件并获得花粉植株;以石刁柏再生植株与栽培植株进行过氧化物酶同工酶电泳鉴别再生植株的性别,其结果表明。
1.石刁柏花蕾的横切面呈蝴蝶形状,由药隔、花粉囊构成。花粉囊包括花粉粒、花药壁二部分。花药壁初期常贮藏大量物质。花药成熟时,细胞径向扩展,贮藏物质消失,除外切向壁外,均发生多条斜纵向条纹状的纤维素次生加厚。花粉囊中的绒毡层的细胞较大,初期为单核的但在花粉母细胞开始减数分裂时,形成双核或多核。花粉囊壁因绒毡层的解体而消失,或仅存痕迹,只剩有表皮及纤维层。

6.石刁柏过氧化物酶同工酶电泳结果,雄株均比相应的雌株少一条酶带。通过与栽培植株过氧化物酶同工酶电泳酶带比较,可以从石刁柏再生植株的过氧化物酶酶带鉴别出性别。
