The aim of this project is: determine the enzymatic activity of alpha amylase with 3.5-Dinitrosalicylic acid colorimetric method (DNS method), then figure out the distribution coefficient K, finally obtain the effect of alpha amylase extraction separation by polyethylene glycol / ammonium sulfate system in the aqueous two-phase system. Through experimental study examines the affection to the distribution coefficient K by the molecular weight of polyethylene glycol, the concentration of ammonium sulfate, pH value, sodium chloride mass fraction, system temperature etc... Thus obtain the best conditions for extraction separation. The research results indicate that alpha amylase extraction effect is better when the mass component of the aqueous two-phase system to be 26% of polyethylene glycol 4000, 21% of ammonium sulfate, 0.002% of sodium chloride, meanwhile pH equal to 5.0 , so the distribution coefficient K can reach up to 3.30.
