Plant material
Open-pollinated seeds of Scots pine, originating from Konginkangas
in central Finland (63° N, 26° E), were surface sterilized

Fungal material
The ECM fungi of Scots pine studied were Pisolithus tinctorius,
originally obtained from the culture collection of the
Swedish University of Agricultural Sciences, Uppsala, Sweden
(Strain 1984a), and Paxillus involutus (Strain H), which
was kindly provided by Prof. V. Hintikka from the University
of Helsinki, Finland. These fungi were chosen because they
differ in their ability to produce IAA (Niemi et al. 2000). Both
fungi were maintained in the culture collection of the University
of Kuopio, Finland, by cultivating the mycelia on
Melin-Norkrans medium (Marx 1969) modified by Heinonen-
Tanski and Holopainen (1991). The medium (MMN1) contained
3.7 mM KH2PO4, 4.7 mM NH4Cl, 0.45 mM CaCl2,
0.43 mM NaCl, 0.61 mM MgSO4⋅ 7H2O, 0.2 μM thiamine-
HCl, 30.8 μM FeCl3⋅ 6H20 and 55.5 mM glucose (pH
5.8), and was solidified with 1.5% agar. Four-week-old fungal
mycelia grown on MMN1 medium were used for studies on
rooting and IAA production.
