简单陈述下,如果我来做这些flow chart的话,每个都包括三个大的部分:
1,模型选取,以第一题为例,是cells isolated from viral induced liver cancers,这里就涉及由于乙肝导致肝癌的病人标本获取(对照组是无肝脏疾病的人),以及从这些标本中分离培养原代细胞。
2.干预手段,以第一题为例,是加入garlic extraction,这里推荐在处理时间和处理量上建立梯度(空白组是只加用于提取garlic extraction的提取液)。
3.检测指标,以第一题为例,on gene expression in cells,这里您得仔细选取去检测哪些基因了,因为讨论的癌症,套路一般是增值、凋亡、迁移,那就选取和增值、凋亡、细胞粘附相关的基因,然后抽rna做RT-PCR,抽蛋白做western。
第二题类似,建模和干预已经在题目里明确了。
第三题就很开放了,我写的话会直接上小鼠,注射小鼠,检测成年鼠和胎鼠,除了肉眼观察有没有畸形外,去五脏六腑肌肉神经做切片并染色,再过分点取血取尿验一系列生化指标,交配个几代看生殖能力,再过分点做点行为学实验看有没有精神病。
第四题和第三题类似,都是看有没有mutagenicity,于是除了对耗子注射物不一样,其他都可以类似。
