建立良好的组织培养体系是进行植物基因转化的基础,研究以西农1338,西农138,西农39,西农133A,西农133B和西农133为材料,对闭颖小麦品种(系)的成熟胚、幼胚和的花药的离体组织培养特性进行了研究,旨在探究不同基因型和外植体对闭颖小麦的愈伤组织的诱导、分化及再生的影响。研究结果表明:闭颖小麦的愈伤组织的诱导与再生与外植体的类型和基因型密切相关。西农133和西农1338的的离体培养特性优良,其中,西农133的成熟胚的培养特性居于首位,达到27.40%(2013)和27.93%(2012),幼胚的培养特性位于第二位,分别为37.75%,西农1338的幼胚的培养特性居于首位,达到40.70%,成熟胚的培养特性位于第二位,达到18.20%(2013)和18.25(2012),西农133和西农1338的花药培养特性结果相同,都为4.48%。总之,这两个品种的实验数据相差不大,与对照材料小偃22处于同一水平。西农133A的离体培养特性最差,明显低于其他供试材料,在所有数据中均位于末位。在3种外植体中,幼胚的培养再生效率最高,成熟胚次之,花药最差,但是供试材料的出愈率与分化率之间不具备明显的相关关系。不同年份收获的成熟胚的离体培养特性比较表明,不同年份的成熟胚的愈伤诱导率有明显差异,但是不同年份的成熟胚的分化成苗率相差不大。

Training system is the basis of plant genetic transformation to establish a good organization, research in Xinong 1338, Xinong 138, Xinong 39,Xinong 133A 133B and Xinong 133, Xinong as materials, the closed glumewheat varieties (lines) of anther and immature embryo and mature embryo,in vitro tissue culture characteristics are studied, influence in order toexplore the different genotypes and explants on closed glume wheat callus induction, differentiation and regeneration. Research results show that:closed glume wheat callus induction type and closely related genotypesand explant and regeneration. Excellent characteristics, the in vitro culture of Xinong 133 and Xinong 1338, Xinong 133 culture characteristic of mature embryos in the first place, to 27.40% (2013) and 27.93% (2012),culture characteristic of immature embryo is second, 37.75% respectively,culture characteristics of Xinong 1338 embryo in the first place, up to 40.70% the culture characteristics of mature embryos, in second place,18.20% (2013) and 18.25 (2012), Xinong 133 and Xinong 1338 anther culture characteristics are the same, is 4.48%. In conclusion, the experimental data of the two varieties were similar, and the control material of Xiaoyan 22 are at the same level. Research of 133A in vitro culture characteristics is the worst, significantly lower than the other testedmaterials, all data are located in the bottom. In 3 kinds of explants,immature embryo culture of mature embryo regeneration efficiency is the highest, the second, anther is the worst, but the tested materials for callus induction rate does not have significant correlation with differentiation rate.Mature embryos harvested in different years in vitro culture shows thecharacteristics of different years, mature embryo callus induction rate have obvious difference, but mature embryos in different years of plantlet differentiation rate difference.
