4. D 此句的Chinese指的是汉语这种语言，是单数，所以很容易就看出答案了。另外must表猜测时，反意疑问句和must无关，只要把前句进行去must的变化后，再进行判断。

Chinese has the largest number of speakers,那么它的反意疑问句当然是doesn't it了。
