2.1. Materials
Seeds of wheat (Triticum aestivum L. var. PBW 502) purchased locally from a seed store wereused in present study. These were surface sterilized with 0.1% (w/v) sodium hypochloritesolution, rinsed thoroughly with distilled water. Cadmium (Cd) treatment wasgiven as cadmium chloride (CdCl2·2(1/2)H2O;molecular weight 228.35) of technical grade (purity = 99%) purchased fromThomas Baker (Chemicals) Ltd., Mumbai, India. Allother chemicals and reagents used in the study for biochemical estimations and enzymaticassay were of reagent grade and procured from Sigma Co., St. Louis, USA;Merck Ltd., India; SiscoResearch Laboratory Pvt. Ltd., India; Acros Organics,Belgium.
2.2. Hydroponicgrowth conditions
Seeds of wheatimbibed for 4 h at room temperature (25◦C) were allowed to germinate on Nylonmesh floating in white plastic trays (23.5 cm × 350px × 7 cm) containing 2 l ofdistilled water. After germination (extrusion of radicle), the trays were placedin a growth chamber set at day/night temperature of 20/10 (±2)◦C, relative humidityof 74–78± 2%, and a photoperiod of 12 h (7.30–19.30) at a photosynthetic photonfluxdensity (PPFD) of approximately 240 [1]molphotons m−2s−1
.The seedlingsgrew for 3 days and when the average root length was nearly 5 cm these weresubjected to different treatments. 展开
