[摘要]目的:采用酸性染料比色法对10批不同产地玛咖样品中总生物碱的含量进行测定,以评价不同产地玛咖的品质。
方法:以氧化苦参碱为对照品,采用紫外-可见分光光度法测定玛咖中总生物碱的含量,并对玛咖总生物碱的提取方法、显色剂的用量、pH值、测定波长进行考察,从而测定不同产地玛咖中总生物碱的含量。
结果:采用超声提取,时间为40min,提取3次,显色剂用量为6ml、pH为7.2、测定波长λ为415nm,经测定玛咖中总生物碱含量相对较高,品质最好的三个地区为秘鲁、丽江和迪庆。
结论:采用本方法测定玛咖中总生物碱的含量及评价玛咖品质操作简单,灵敏度高、重现性好。 展开

Abstract:
Objective- To determine the total alkaloid in 10 batches of Maca samples from different regions by adopting the acid dye colorimetric method, and to evaluate the maca quality of different regions.
Methodology - By using Oxymatrine as a reference substance, UV-VIS spectrophotometry was adopted to determine the total alkaloid contents, and the extraction method, the dosage of chromogenic agent, pH values, the measured wavelengths were observed, and the total alkaloid of maca from different regions were determined.
Result - 3 extractions were conducted by applying the method of ultrasonic extraction, and with 6 ml of chromogenic agent, 7.2 pH value, 415nm measured wavelength, the total alkaloid in the maca samples were found to be relatively high,
and the three best maca quality regions were Peru, Lijiang and Diqing.
Conclusion - It was found that the advantages of using this method to determine the total alkaloid in maca and evaluate the quality were simple operation, high detection sensitivity and good reproducibility.
