In order to make genome organization of Betula luminifera BISPL2, provide basis for its SNP analysis, this research according to cDNA sequence designs pecullar primer, takes total DNA of Betula luminifera genome as template to conduct PCR amplificaton and sequencing analysis, and compare with cDNA sequence. The result shows that overall length of BISPL2 genome is 3554bp, it contains 2 intron, 3 exon. Length of the first intron is 1005bp, of the second is 624bp. Length of the 3 exon are 416, 134 and 620bp, respectively. At the third exon (3025-3044) exists miR156 locus, length of 5’-UTR is 485bp and of 3’-UTR is 169bp. On this basis, for its second intro and 3’-UTR area about 850bp, designs primer clone gene segment used for SNP analysis, clone corresponding segment taking 75 clones from different provenance as materials. Using MEGA5.0 to compare and analyze sequence of 75 genetype indivudual of Betula luminifera. By using of DNsp5, detect 103 single nucleotide polymorphisms. SNP frequency is 1/8bp. Of these SNPs, 96 are dimor-phism SNPs, among which 52 blong to transition, 44 belong to transversion. The result of linkage disequilibrim analysis to SNPs in SPL2 gene shows that along with the increase of nucleotide sequence length, linkage disequilibrim of SNPs grandually increases in the gene, but gently. The research results provide theoretical basis for linkage disequilibrim mapping of Betula luminifera SPL gene and its genes assisting Betula luminifera control breeding of flowering molecular.
