These reports described that mice fed on a high-fat diet for 2 to 4 weeks exhibited a significant increase in plasma LPS. This increase in LPS in the plasma induced by a high-fat diet suggests metabolic endotoxemia, Which is considered to trigger the development of obesity , inflammation , insulin resistance , type 2 diabetes and atherosclerosis via activation of the CD14/TLR4 receptor complex by LPS and/or the fatty acids. Furthermore , it has also been demonstrated that the population levels of Bifidobacterium spp . and the Eubacterium rectal /clostridium coccoides group were significantly reduced in high-fat-fed mice as compared to those receiving the standard high carbohydrate diet .The administration of oligofructose (OFS) to high-fat-fed mice may thus abolish the increase in plasma LPS With associated decreases in energy intake , body Weight gain ,glycemia and epididymal fat mass. The Bifidobacterium spp.levela were significantly increased in OFS-administered mice , while the plasma LPS levels decreased.
All these findings strongly suggest that gut microbiota contribute to the complications related to high-fat diet feeding . it would thus be useful to develop specific strategies using prebiotics for modifying gut microbiota to favor specific gut microbiota and to prevent the delirious effects of high-fat or obesity-induced complications. Bifidobacteria are gram-positive anaerobic microorganisms that naturally colonize the human intestinal tract and are believed to be beneficial to human health . Several previous studies have shown that bifidobacteria lowered the intestinal endotoxin level and improved the mucosal barrier function . However, the effect ot administering specific probiotics added to high – fat diet-induced obesity has not been defined . we evaluated in the present study the effect of an oral administration of the bifidobacterial strain ,Bifidobacterium breve B-3, in a mouse model with high-fat diet –induced obesity and insulin resistance.
