2.造模成功后选取符合研究标准的大鼠32只进行分组,分为B组:模型组8只;C组:贝那普利组8只;D组:中药组8只;E组:益肾汤联合贝那普利组简称中西医结合组8只,加上A组共5组。
3. 造模成功后即开始给药,各组分别给予相应药物的水煎剂。
4. 实验结束后检测24小时尿蛋白定量及血BUN、Scr 、RBC、Hb,并进行肾组织学观察和肾组织中TGF-β1 表达检测。
结果:
1.一般情况:实验期间,假手术组大鼠表现机警,反应快,皮毛致密、整齐而有光泽,生长、进食及活动情况均无明显异常,体重增加;模型组明显营养不良,精神萎靡,活动迟缓,食欲不振,皮毛蓬松、枯槁无光泽,在治疗过程中死亡2只,考虑可能由于肾衰竭所致;贝那普利组大鼠精神较假手术组萎靡,活动慢,皮毛蓬松;中药组大鼠表现与贝那普利组类似;中西结合组大鼠与假手术组大鼠一般表现无太大差别,只是皮毛干燥无光。

结论:益肾汤通过益气健脾、活血化瘀、利湿泻浊,能降低CRF大鼠血BUN、Scr,改善贫血,降低蛋白尿,能有效降低5/6肾切除致CRF大鼠肾组织中TGF-β1 的表达,从而减少ECM 的积聚,减缓肾纤维化的发展,延缓CRF的进展。充分说明益肾汤具有良好的抗肾纤维化作用,而且联合贝那普利后疗效更佳。
